The advancement in research has granted humans an understanding of how living organisms function and the purpose of their function: how they interact with the environment, the type of inter-species interactions that they have, and the impact such interactions have on the rest of the planet. These explanations have given rise to further questions based on the origin and evolution of all biological species, which eventually leads to queries regarding the exact processes occurring within an organism that determine all aspects of its existence, including the impact it has on its environment.

At present, the rising progress in science, medicine, and technology has made positive as well as negative impacts on the planet, as a whole. Current scientific research hopes to resolve these negative impacts with biological solutions, while at the same time demystifying and resolving queries related to the biota.

Aging process

Aging or senescence is the collective process through which an organism undergoes multiple physiological changes over the course of its lifetime. It includes the development of an organism’s cognitive functions till it reaches maturity, and then a steady decline of those same functions till the organism’s eventual death. Such a decline affects other body processes, leading to their impaired functioning.

In humans, aging brings with it, body frailty, organ weakness, and mental decline. Research has shown that humans are born with a biological clock that regulates this process of aging. This clock functions by the combination of a steady loss of hexamer telomeric repeat sequences of chromosomes during replicative processes and the accumulation of DNA damage and cellular wastes in an organism during the course of its life.

Scientists have researched model organisms such as C. Elegans, and observed reduced senescence in the event of selective mutations in specific genes. However the same result is yet to be reproduced in higher and more complex organisms, hence the elucidation of the exact process remains a mystery. As more studies are carried out, more theories are put forth to help explain the occurrence of this process, along with ways to prevent and reverse it.

Agriculture

The ever-growing human population along with the recent population explosion has caused a dramatic increase in the demand for food crops. This coupled with the occurrence of natural disasters like earthquakes, floods, etc., and pest problems, result in huge losses of food grains. Such a shortage increases the world’s hunger problem, where most of the inhabiting people of developing nations are unable to feed themselves everyday.

To solve such a mammoth crisis, and to avert death of people on account of starvation, the yield of food has to be increased manifold. One way to do this, can be by upgrading the agricultural practices of farmers; by giving them advanced know-how, better tools and implements, and to have precautions in place to protect the farmed yield from instances of natural disasters and pest populations.

Another way to overcome this problem, is to research and produce newer varieties of these food crops that are resistant and even repellent towards pest populations, while at the same time produce a very high yield with minimum care. Varieties of crops which produce food in more than one season, can also be produced.

Antibiotic Resistance

With the advent of vaccines, anti-microbial, and anti-viral agents, numerous diseases have been eradicated, averted and contained. Infections that would have proven fatal a few decades ago are now taken care of by a mere injection, tablet, or a capsule, helping in increasing the overall health of the human population.

But with the excessive use of antimicrobial solutions, nature serves us a reminder of continuous evolution in the form of antibiotic resistant strains of micro-organisms. As the strength and efficiency of these antibiotics evolves due to medical research, the infectious organisms too evolve and adapt to overcome these substances. An infection caused by such an organism is not only untreatable, but also proves to be fatal.

To curb this evolution of “super-bugs”, newer mechanisms and pathways have to be researched to prevent infections by these organisms. Also, targeted drugs have to be developed that halt the organism’s internal processes, thereby rendering it unable to infect.

Technology has brought the world closer and improved the quality of life considerably. Actions that took extended periods of time, can now be carried out easily and within a short time. There is year round availability of goods in all parts of the world.

But such industrial advancement comes with its fair share of adverse consequences. The petroleum industry which provides fuel for so many purposes, has accidents which cause oil spills in oceans. These affect the natural fauna and flora of the marine ecosystem. The smoke emissions from vehicles and industries cause air pollution, which in turn affects the health of animals and humans equally. The effluents from the factories pollute water bodies, that eventually affect anything and anyone that utilizes the water.

These instances are not only hampering the health of organisms but also affecting the physical world in the form of climate change, global warming, and ozone depletion. Scientists are now working on natural biological ways to rid the environment of such harmful pollutions, and are also devising ways in which the pollution is altered and reduced at their very source.

Cancer Metastasis

These cells have the potential to spread to other parts of the body, and spread the cancer. Cancer is characterized by the formation of a cell mass called the tumor. There are two types of tumors – benign and metastatic. The benign tumor is just a compact overgrowth of cells that can be easily removed by surgery. However, if the cells turn metastatic, that is they gain the ability to migrate to other parts of the body via the blood stream, the tumor is said to be malignant.

Maximum cases of death due to cancer are due to recurrences of the tumor. Tumors once removed, recur due to the metastatic nature of the cells. If the metastasis is prevented or averted, the chances of the tumor recurring are drastically reduced.

Not only is this condition without a cure, but the mechanisms by which it occurs is also not known in detail. Research has to be carried out to determine the mechanism by which it occurs, and only then could a way be worked out to prevent and eventually cure this condition.

All organisms develop from a single-celled zygote, and animals within the same order and family of classification resemble each other during the initial stages of embryonic development. The field of developmental biology deals with the study of how complex multicellular organisms develop and emerge from what is essentially a ball-like mass of cells (morula).

It consists of studying the various processes that give rise to our characteristic physical appearance, body plan and structures, and the regulatory processes of the body. Any defect or malfunction during the embryogenesis process causes a structural and functional defect in the individual organism. In some cases the defect is too large to overcome, and proves fatal to the organism, leading to the death of that developing embryo.

Developmental defects cause a variety of health disorders such as spina bifida, microcephaly, schistocystis, etc. Researching the causes of these conditions may offer an insight into their management and possible cure.

Any disorder that occurs, is mainly due to a mutation or defect in the gene sequence. This disruption causes a change in the protein encoded by that gene. The altered protein leads to a malfunction or prevents the occurrence of a process eventually, thereby giving rise to the disorder.

A few such disorders include Downs syndrome, Huntington’s disease, Marfan syndrome, Alzheimer’s, Lesch-Nyhan syndrome, and Cystic Fibrosis. Studying the mechanisms of these diseases will help elucidate the path along which the disease progresses, and the role of the genes involved.

Hallmarks of precancer: Researchers investigate the principles governing the biology of early, precancerous lesions

Every oncologist and cancer researcher is familiar with the hallmarks of cancer, a series of functional capabilities that human cells acquire as they transition from a normal state to a neoplastic state (a state of excessive and abnormal growth). These hallmarks have been used (and updated) during the last quarter of a century as a "conceptual scaffold" to help "rationalize the complex phenotypes of diverse human tumor types and variants in terms of a common set of underlying cellular parameters."

Now, Ken Lau, professor of cell and developmental biology , and Neelendu Dey, assistant professor of medicine at the University of Washington, and colleagues have laid out the principles governing the biology of early, precancerous lesions , which are different from the principles that govern cancers. Their authoritative perspective was published in Cancer Discovery in April 2024.

A large body of research details how lifestyle factors can predispose a person to cancer initiation and development, but how those macroenvironmental risk factors are manifested in cells and molecules is poorly understood. Lau, Dey, and colleagues shed light on these points.

"Precancers are defined as 'at-risk tissues and lesions' that develop into cancer," Lau said. "Given the effectiveness of early interventions against cancers, we hope to establish some principles that the research community can follow when looking at precancers."

These principles include age-related genetic alterations, epigenetic changes, metabolic alterations, the hijacking of regenerative cell state transitions, the disruption of immune surveillance and "inflammaging," and remodeling of the tissue microenvironment mediated by senescence or biological aging. Inflammaging refers to the state of chronic, sterile, and low-grade inflammation that naturally occurs with advanced age.

"By understanding the hallmarks of precancer, we hope that we can further develop early detection, stratification, and intervention strategies at the precancer stage," Lau said.

Just as the hallmarks of cancer are updated as we develop new knowledge, Lau and colleagues hope to update the hallmarks of precancer as we learn more about them.

Environmental Changes Are Fueling Human, Animal and Plant Diseases, Study Finds

Biodiversity loss, global warming, pollution and the spread of invasive species are making infectious diseases more dangerous to organisms around the world.

Several large-scale, human-driven changes to the planet — including climate change, the loss of biodiversity and the spread of invasive species — are making infectious diseases more dangerous to people, animals and plants, according to a new study.

Scientists have documented these effects before in more targeted studies that have focused on specific diseases and ecosystems. For instance, they have found that a warming climate may be helping malaria expand in Africa and that a decline in wildlife diversity may be boosting Lyme disease cases in North America.

But the new research, a meta-analysis of nearly 1,000 previous studies, suggests that these patterns are relatively consistent around the globe and across the tree of life.

“It’s a big step forward in the science,” said Colin Carlson, a biologist at Georgetown University, who was not an author of the new analysis. “This paper is one of the strongest pieces of evidence that I think has been published that shows how important it is health systems start getting ready to exist in a world with climate change, with biodiversity loss.”

In what is likely to come as a more surprising finding, the researchers also found that urbanization decreased the risk of infectious disease.

The new analysis, which was published in Nature on Wednesday, focused on five “global change drivers” that are altering ecosystems across the planet: biodiversity change, climate change, chemical pollution, the introduction of nonnative species and habitat loss or change.

The researchers compiled data from scientific papers that examined how at least one of these factors affected various infectious-disease outcomes, such as severity or prevalence. The final data set included nearly 3,000 observations on disease risks for humans, animals and plants on every continent except for Antarctica.

The researchers found that, across the board, four of the five trends they studied — biodiversity change, the introduction of new species, climate change and chemical pollution — tended to increase disease risk.

“It means that we’re likely picking up general biological patterns,” said Jason Rohr, an infectious disease ecologist at the University of Notre Dame and senior author of the study. “It suggests that there are similar sorts of mechanisms and processes that are likely occurring in plants, animals and humans.”

The loss of biodiversity played an especially large role in driving up disease risk, the researchers found. Many scientists have posited that biodiversity can protect against disease through a phenomenon known as the dilution effect.

The theory holds that parasites and pathogens, which rely on having abundant hosts in order to survive, will evolve to favor species that are common, rather than those that are rare, Dr. Rohr said. And as biodiversity declines, rare species tend to disappear first. “That means that the species that remain are the competent ones, the ones that are really good at transmitting disease,” he said.

Lyme disease is one oft-cited example. White-footed mice, which are the primary reservoir for the disease, have become more dominant on the landscape, as other rarer mammals have disappeared, Dr. Rohr said. That shift may partly explain why Lyme disease rates have risen in the United States. (The extent to which the dilution effect contributes to Lyme disease risk has been the subject of debate, and other factors, including climate change, are likely to be at play as well.)

Other environmental changes could amplify disease risks in a wide variety of ways. For instance, introduced species can bring new pathogens with them, and chemical pollution can stress organisms’ immune systems. Climate change can alter animal movements and habitats, bringing new species into contact and allowing them to swap pathogens .

Notably, the fifth global environmental change that the researchers studied — habitat loss or change — appeared to reduce disease risk. At first glance, the findings might appear to be at odds with previous studies, which have shown that deforestation can increase the risk of diseases ranging from malaria to Ebola. But the overall trend toward reduced risk was driven by one specific type of habitat change: increasing urbanization.

The reason may be that urban areas often have better sanitation and public health infrastructure than rural ones — or simply because there are fewer plants and animals to serve as disease hosts in urban areas. The lack of plant and animal life is “not a good thing,” Dr. Carlson said. “And it also doesn’t mean that the animals that are in the cities are healthier.”

And the new study does not negate the idea that forest loss can fuel disease; instead, deforestation increases risk in some circumstances and reduces it in others, Dr. Rohr said.

Indeed, although this kind of meta-analysis is valuable for revealing broad patterns, it can obscure some of the nuances and exceptions that are important for managing specific diseases and ecosystems, Dr. Carlson noted.

Moreover, most of the studies included in the analysis examined just a single global change drive. But, in the real world, organisms are contending with many of these stressors simultaneously. “The next step is to better understand the connections among them,” Dr. Rohr said.

New research published in Current Biology sheds light on how animals create and maintain internal spatial maps based on their surroundings.

The study, led by Dr. Guifen Chen from Queen Mary University of London, delves into the brains of mice navigating a two-dimensional virtual reality (VR) environment, revealing the surprising importance of specific visual cues for building and maintaining spatial maps. It reveals that specific visual cues—in this case, elevated walls—are crucial for stabilizing the neurons responsible for spatial navigation in virtual reality (VR).

"Our findings provide a significant step forward in understanding the precise nature of the sensory information that animals used for boundary detection," says Dr. Chen. "They not only highlight the importance of elevated boundaries in building spatial maps, but also reveal the brain's remarkable ability to infer boundaries from sensorimotor mismatch even when they're not directly visible."

The research team conducted a fascinating experiment using virtual reality techniques. Mice navigated in a two-dimensional virtual environment, while the neural activity was monitored. Specifically, the study focuses on the activity of neurons crucial for navigation: place cells, which fire when the animal is in a specific location, and grid cells , which form a hexagonal grid-like map of the environment.

This VR environment was a two-dimensional world that could be manipulated to include or exclude various visual elements. By monitoring the activity of these neurons, the scientists could observe how the mice's spatial maps were updated in response to the manipulation within the VR world.

The most striking finding centered around the role of visual boundaries. When the VR environment included elevated walls, the place cells and grid cells in the mice's brains fired consistently, indicating stable spatial maps.

However, removing these walls caused the firing patterns of these cells to become erratic, demonstrating a disruption in the animals' ability to navigate. Interestingly, removing cues from the floor of the VR environment had no significant impact. This suggests that the specific form of visual cues plays a crucial role in how animals build and maintain their internal maps.

Dr. Chen worked with Xiuting Yang, a Ph.D. student in her lab at the School of Biological and Behavioural Sciences at Queen Mary University of London, as well as Professor Francesca Cacucci, Professor Neil Burgess and Dr. Tom Wills at UCL on this paper.

The research team believes these findings have broader implications for understanding real-world navigation.

"Our results suggest that the elevated—not flat—boundary plays a crucial role in how animals maintain spatial maps," explains Dr. Chen. "This may explain why, for instance, young children struggle to use flat outlines of shapes for spatial orientation."

This study opens doors for further research into the intricate interplay between sensory information, spatial memory, and navigation. It could pave the way for advancements in fields ranging from robotics and virtual reality development to a deeper understanding of spatial navigation disorders.

Promising new treatment strategy for deadly flu-related brain disorders

It might start simply, with a cough or sneeze…but in some cases, the flu virus can make its way to your brain, causing serious symptoms or even death through a disease known as influenza-associated encephalopathy (IAE).

Now, in a study published in Acta Neuropathologica , researchers have revealed that IAE may be caused by the virus entering the brain through a specific cell type, and have identified possible treatment strategies.

Although IAE is increasingly common, surprisingly little is known about how the influenza virus actually gets into the brain and causes symptoms of encephalopathy (a general term for brain disorders). Notably, a definitive treatment for IAE remains lacking, something that researchers from Osaka University wanted to address.

To study how influenza viruses might cause IAE, the research team used a range of methods. As well as looking for virus particles in the brains of people who died from IAE, they created a mouse model of the disease by injecting influenza A virus into the blood. They also used cell culture methods to look at how well the virus infects different cell types.

"In the human brains, the virus-injected mice, and the cultured cells, the influenza virus tended to accumulate in endothelial cells," explains lead author of the study Shihoko Kimura-Ohba. "These cells create a barrier between the blood and the brain, and are important for protecting the brain from harmful substances."

Accordingly, in both the human brains and the mouse model, the barrier between the blood and the brain was compromised. Furthermore, the researchers noticed that the virus wasn't actually reproducing within these endothelial cells -- but there were a lot of proteins made by the virus.

"When we saw this viral protein accumulation in the brain, we realized that antiviral medications aimed at stopping viral proliferation were unlikely to help," says Tomonori Kimura, senior author of the study. "However, antivirals aimed at stopping viral transcription and translation -- vital processes for making proteins -- are also available."

When the team tried these classes of antivirals in endothelial cells treated with influenza virus, they observed less viral proteins and cell death. In the mouse model of IAE, these antivirals were also very effective when used early enough, suggesting that they may be useful in human patients.

Given that many different kinds of viruses, including COVID-19, can cause encephalopathy, these findings have wide-ranging impacts. Moreover, although IAE care is improving, more than half of all patients die or have long-lasting symptoms. A better understanding of how viruses can cause encephalopathy will be important for developing new, effective treatments.

Biointelligence and Biosecurity for the Intelligence Community (B24IC) program seeks to develop new capabilities, matching the wider synthetic biology and biotechnology fields, ensuring the Intelligence Community’s (IC’s) capability to meet the biointelligence and biosecurity threats of the 21st century. This includes developing new ways to collect, detect, analyze, and prevent traditional biothreats while also addressing the promise and perils associated with the growing fields of biotechnology and synthetic biology. To address these challenges, the IC seeks to advance research across multiple sub-disciplines of biology.

In recent decades, the rise of synthetic biology has corresponded with critical advances in biology research. From polymerase chain reaction (PCR) in the 1980s to next generation sequencing (NGS) in the late 90s to numerous mechanisms for genetic engineering enabling a variety of engineered organisms in the past decade, the branches of synthetic biology and enabling biotechnologies have advanced at a prodigious rate. The IC, and IARPA specifically, has pursued numerous research programs to advance security interests, but the needs of the IC require rapid advancement of numerous research topics to meet and leverage the advances the many biological disciplines have brought into reality in the past decades. These needs align with developing new methods for countering traditional biothreats of concern while also looking towards the future where bio-focused capabilities may enable or support IC relevant capabilities.

Research Topics

Gametes, such as sperm and eggs, are produced through meiosis. In this specialized type of cell division, a single round of DNA replication is followed by two successive rounds of chromosome segregation. In the first division, homologs (each parental chromosome) separate; in the second division, the sister chromatids segregate, resulting in four haploid daughter cells. The segregation of homologs poses a unique challenge because they must be physically connected for accurate segregation. The connection between homologs, in most mammals, is mediated by the deliberate introduction of hundreds of double-stranded breaks (DSBs) and their subsequent repair through homologous recombination. The cell needs to fine-tune the timing and placement of DSBs to avoid deleterious events, such as mutations or aneuploidies. 20% to 60% of human oocytes have a karyotypic defect, and errors in meiosis are responsible for at least half of clinically recognized miscarriages, as well as a spectrum of chromosomal birth defects in humans, such as Down syndrome.

The main focus of our research is to understand the multiple layers that shape the recombination landscape over size scales ranging from single nucleotides to whole chromosomes in the human and mouse germlines. To achieve this goal we use a combination of genetics, multi-omics analysis and microscopy. Successful meiosis requires the coordination of multiple cellular and molecular processes. Through this research program, I seek to advance our knowledge of a critical biological process at the core of modern genetics that profoundly affects human reproductive health and disease.
