Who you are You are an ambitious master student who is currently enrolled in Toxicology, Biology, Biochemistry or comparable. You are a phenomenal teammate, motivated by meeting early drug discovery, patient as well as drug safety and you would like to learn more about how to address challenging questions experimentally. Master´s thesis – Cell culture medium development in Gene Therapy

The Position

The cell culture medium is one of the most critical parts of the cell culture process because of its impact on cell growth, productivity, product quality and process robustness. The Gene Therapy Cell Culture Technical Development Team at Roche is seeking a motivated student to directly contribute to cell culture medium and platform development efforts through the development of medium feed supplements and design of an optimal feeding strategy. Your work will be an integral part of our global platform development strategy.

Key activities and responsibilities

Leveraging past studies and extracting critical information from the scientific community to develop an understanding for cellular metabolism and requirements

Developing a panel of medium feed supplements based on e.g. limiting metabolites obtained from spent medium analytics

Designing different feeding strategies for those supplements to optimize cell growth, AAVv production and product quality

Performing growth and AAVv production experiments in laboratory-scale (shake flasks or shaken multiwell plates) and verifying results in controlled bioreactors

Statistical data analysis and interpretation of obtained results

Working both individually and as part of a team to achieve project goals within defined timelines

Results of the project will be presented and discussed in a global team

Who you are

You are currently enrolled in a master’s course in biotechnology, biology, biochemistry, process engineering or a comparable course

Experience in cultivation of mammalian cells is very preferred

Knowledge of metabolism of mammalian cell cultures, e.g. HEK293, composition of cell culture media, virology, cultivation systems and strategies, and statistical data analysis would be an asset

You are characterized by your careful, independent and responsible way of working as well as your enjoyment of working in a team

You are proficient in English and confident in communicating withinternational colleagues in person, via phone or Email.

Start from summer 2024 (earliest September) with a three to six-month internship and a subsequent six-month master´s thesis.

Was Sie erwartet:

In der Abteilung "Global Engineering – Hardware Systems Engineering" bei Roche Diabetes Care werden unter Beachtung aller relevanten Richtlinien Insulinpumpen, Blutzuckermessgeräte und Stechhilfen sowie die dazugehörigen Applikationssysteme (z.B. Teststände) serienreif entwickelt.

Im Rahmen einer Bachelor- oder Masterarbeit sollen Simulationsmodelle aufgebaut und die Wiederverwendbarkeit bewertet werden. Die Simulationen haben sich im Entwicklungsprozess etabliert und bieten viele Vorteile. Zunehmend können Versuchsaufwände und auch die Entwicklungszeit durch den Einsatz von Simulationswerkzeugen gesenkt werden. Der Wert von Simulationsmodellen kann erheblich gesteigert werden, wenn die Wiederverwendung steigt. Hierbei sind Schnittstellen zu anderen Simulationswerkzeugen ein wesentlicher Schritt, um einen digitalen Zwilling (digital twin) zu ermöglichen oder Metamodelle abzuleiten. Die gleichen Modelle, oder Komponenten dieser, können idealerweise unterschiedlichen Simulationswerkzeugen zugänglich sein und somit problemspezifisch genutzt werden.

Folgende Schwerpunkte stellen dabei Ihre Aufgabe dar:

Ermittlung des Standes der Technik

Sammeln von Anforderungen und Ableitung von erforderlichen Eigenschaften eines Frameworks

Erarbeitung methodischer Ansätze

Aufbau eines Simulationsmodells als Teil des Gesamtmodells, beispielweise des Antriebsstrangs der Insulinpumpe

Bewertung der methodischen Ansätze

Definition eines Frameworks für Simulationsmodelle

Implementierung des Frameworks anhand von Simulationsmodellen

Bewertung der Testimplementierung

Vorstellung der Ergebnisse

Dokumentation

Wer Sie sind:

Sie studieren in der Fachrichtung Elektrotechnik, Mechatronik, Maschinenbau, Mikrosystemtechnik, Medizintechnik oder vergleichbares und wollen Ihre Bachelor- oder Masterarbeit im einem spannenden Umfeld durchführen.

Sie besitzen grundlegende Kenntnisse in der Simulation im technisch physikalischen Umfeld.

Sie besitzen eine schnelle Auffassungsgabe und begeistern sich für methodische Frameworks sowie schlanke Prozesse.

Sie besitzen erste Erfahrung in der Programmierung mit Matlab, Python oder einer ähnlichen Skriptsprache.

Kenntnisse in SysML und Enterprise Architect sind von Vorteil.

Sie freuen sich darauf sich in Teamstrukturen zu integrieren.
